Great video and informative. Sleep deprivation, chronic inflammation, chronic invasive infections (especially intracellular infections), low to deficient Vitamin D3, Inactivation of Brown Fat and certain nutritional deficiencies are the main drivers of mitochondria dysfunction. <br><br>The major inflammatory markers are IL-6, TNF-a, IL-1b and others along with pathways like NFKB. Cortisol is released in response to chronic NFKB inflammation in an attempt to lower the inflammation pathways and lower the inflammatory markers like IL-6. <br><br>IL-6, TNF-a are 2 of the main drivers of mitochondrial inflammation or dysregulation. Low vitamin d3 and low to deficient estrogen increases this inflammation creating high cell stress and elevated or uncontrollable ROS which affects mitochondrial function or causes mitochondrial dysfunction. Estrogen is more supportive for cell function than being a causative agent. <br><br>In addition, due to the high work load of mitochondria, the body’s normal functioning is going to have damaged and or underperforming mitochondria that need to be replaced hourly on a daily basis. Sometimes just the mitochondria need to be replaced (Mitophagy) and sometimes due to so much damage or uncontrollable ROS, the mitochondria send signals to institute cell apoptosis. Mitochondria dysfunction begins to develop when these two processes break down due to high inflammatory states. In other words, your immune system is supposed to be able to handle this important process and replace damaged or non functioning mitochondria. <br><br>To illustrate the aforementioned, every human including healthy humans create dysfunctional cells daily (cancer cells or precancer cells). Yes, your body makes about 10,000 cancer cells per day. Your immune system is designed to identify and eliminate those cells through various processes including apoptosis pathways like Ferroptosis. When your immune system is compromised, the cancer cells can’t be destroyed and cancer develops. Cancer is the result of immune system dysregulation. <br><br>Just like cancer, if your cell can’t rid itself of damaged mitochondria, the cell accumulates ROS and damaged mitochondria. The mitochondria develop increased Fission and lowers Fusion. This imbalance is one of the main factors in causing mitochondrial dysfunction. <br><br>Basically, mitochondrial damage occurring faster than mitochondrial quality-control systems can repair, remove, and replace the damaged mitochondria becomes the main problem. <br><br>Vitamin D3 is a component of the autophagy and apoptosis pathways. So low levels of vitamin d3 can hinder healthy functioning. <br><br>The main things that elevate IL-6 is HFCS, Ultra Processed Foods, Chronic invasive infections like staphylococcus aureus, viruses, poor sleep, inactivated Brown Fat. There are many others, but these are some of the main issues. Interestingly, new research shows the immune system actually begins to create visceral fat depots when patients begin to become infected with staphylococcus aureus. I have a Post on my channel explaining this including supported with clinical studies. 80% of all patients having been diagnosed with mold toxicity are also diagnosed with staphylococcus aureus infection in their nasal passages (Richard Shoemaker MD 10,000 patients). <br><br>To lower IL-6 you need to eat Whole Foods and avoid UPFs. You should exercise properly and get your vitamin d3 levels to around 90-110 (high end of normal range). <br><br>DHA and EPA may also help support mitochondrial function and quality control. These fatty acids become incorporated into mitochondrial membrane phospholipids, where they influence membrane properties and interactions with energy-producing proteins. Human research shows that EPA/DHA supplementation can improve how mitochondria respond to ADP—the signal to produce ATP.<br><br>Experimental studies also show that DHA can promote the removal of damaged mitochondria through PINK1–Parkin-associated mitophagy and reduce excessive mitochondrial fragmentation under inflammatory stress. Healthy mitochondria need coordinated fission, fusion, and clearance. Sometimes fission must increase to separate damaged sections for removal; in other situations, excessive fragmentation needs to decrease. The goal is effective mitochondrial maintenance. <br><br>Other nutritional deficiencies can impair the systems that maintain mitochondrial health. Choline is needed to produce phosphatidylcholine, a major membrane phospholipid. Vitamin E helps protect membrane lipids against oxidative damage. Magnesium supports oxidative phosphorylation and reactions involving ATP. These nutrients contribute to membrane integrity, protection, and energy production through different mechanisms. <br><br>The B vitamins are important too. Vitamin B1—thiamine—helps enzymes process fuel for energy production. Vitamin B2—riboflavin—provides the FAD and FMN cofactors used in energy-producing reactions. Vitamin B3—niacin—provides NAD and NADP, which support energy metabolism and cellular antioxidant systems. Deficiencies can therefore interfere with the reactions mitochondria depend on to function properly. <br><br>Vitamin D signaling also influences mitochondrial function and quality-control pathways. Experimental research links vitamin D receptor activity with proteins involved in mitophagy, including PINK1 and BNIP3. Correcting nutritional deficiencies, obtaining sufficient sleep, and exercising regularly can support mitochondrial health. Direct evidence that DHA/EPA supplementation restores healthy mitophagy in humans is still limited. Eating fatty fish like salmon and Sardines are a great way to get the fatty acids & pharmaceutical grade levels of DHA/EPA with proper Fatty Fish diets. I just posted a Fatty Fish/Sardine diet explaining all the different pathways said diet activates which makes the Fatty Fish/Sardine diet one of the most effective weightloss diets you can eat. The diet stimulates 14 different Brown Fat Activating pathways I have listed in the post.

Fasting works fine for mitochondria repair through mitophagy and it's all for free. I don't think it's necessary to waste money on supplements to get a similar result!!!!!

Thomas, I so much appreciate and can relate to your passion and drive to learn and better understand these biological processes, like autophagy and mitophagy. I'm a bit different in that I'm desperate to find solutions to help heal and strengthen chronic severe injuries that have greatly limited my mobility, and life experience in general. Regardless, I think I have benefitted from your own activities of study and data presentations. While I'll never be at your level, I'm still hoping and striving with everything I've got, for significant improvement - relative to my own status and for as long as it takes. So thanks for continuing to deliver the ideas, info and passion.

More importantly, do something on the ratio of D3 versus K2 and how it is killing so many people by pushing calcium into their blood by being the wrong ratio and also high protein! Especially for those over 50.
